Clostilbegyt is a drug with an antiestrogenic effect.

Release form and composition

Clostilbegyt is produced in the form of tablets: white, grayish-white or yellowish-white, flat, round, beveled, with the inscription "CLO" on one side, without or almost odorless (10 pcs. The composition of 1 tablet includes:
    Active ingredient: clomiphene citrate - 50 mg;
    Auxiliary components: gelatin - 2 mg, magnesium stearate - 2 mg, stearic acid - 2 mg, talc - 5 mg, potato starch - 39 mg, lactose monohydrate - 100 mg.

Pharmacodynamics

The active ingredient of Clostilbegyt is clomiphene - an antiestrogenic agent of a non-steroidal structure.
The mechanism of its action is explained by the ability to specifically bind to estrogen receptors of the pituitary gland and ovaries. In patients with a low estrogen content in the body, the drug exhibits a moderate estrogenic effect, in patients with a high estrogen content - an antiestrogenic effect.
When used in small doses, it enhances the secretion of gonadotropins (follicle-stimulating hormone, luteinizing hormone and prolactin), stimulates ovulation. In high doses, clomiphene inhibits the secretion of gonadotropins.
Clostilbegyt does not possess androgenic and gestagenic activity.

After oral administration, clomiphene is well absorbed from the gastrointestinal tract.

Instructions for use: method and dosage

In the treatment of infertility, the dose and duration of the treatment course are determined by the sensitivity to the action of Clostilbegyt ovaries.
It is recommended to start treatment for women with a regular menstrual cycle on the fifth day of the cycle or on the third day of the cycle if the follicular phase lasts less than 12 days or early ovulation. With amenorrhea, therapy can be started any day.
First, Clostilbegyt tablets are used according to the following scheme: for 5 days, 50 mg per day with simultaneous control of the ovarian reaction using laboratory and clinical studies. As a rule, ovulation occurs between 11 and 15 days of the cycle.
If therapy does not lead to ovulation, the drug is used according to the second scheme: for 5 days, starting from the 5th day of the next cycle, take 100 mg of Clostilbegyt per day. If ovulation does not occur at this time, the same scheme must be repeated again. With continued anovulation, you should take a three-month break in taking the drug, and then repeat the therapy within the next 3 months. If the repeated course is ineffective, further use of Clostilbegyt is not effective. For a maximum of each course, you can take 750 mg of the drug.
The initial daily dose of the drug for polycystic ovary syndrome is 50 mg (due to the tendency to hyperstimulation).
With post-contraceptive amenorrhea, Clostilbegyt is taken daily at 50 mg. In most cases, a five-day course of the first treatment regimen described is successful.
In accordance with the indications for men, the drug is prescribed 1-2 times a day, 50 mg. The duration of therapy is 6 weeks (under systematic sperm analysis).

Side effects

During the use of Clostilbegyt, the following side effects may develop (> 1% - often; <1% - rarely):
    Endocrine system: dysmenorrhea, breast induration, abnormal uterine bleeding, an increase in the size of the ovary (including cystic); rarely - soreness in the area of ​​the mammary glands. During therapy, the likelihood of endometriosis, ectopic pregnancy, multiple pregnancy, and the growth of existing uterine fibroids increases. Cystic enlargement of the ovaries is also possible, especially with Stein-Leventhal syndrome (the size of the ovaries can reach 4-8 cm). In this case, you need to monitor the body temperature and, as soon as it becomes biphasic, interrupt therapy;
    Central nervous system: headache, drowsiness, dizziness; rarely - slowing down of motor and mental reactions, depression, hyperexcitability, insomnia;
    Digestive system: vomiting, nausea; rarely - acute abdominal syndrome, flatulence, gastralgia, diarrhea, increased appetite;
    Reproductive system: vaginal dryness, pain in the lower abdomen;
    Urinary system: polyuria, increased urination;
    Sense organs: visual impairment (including blurred contours, double vision, impaired perception of light, photophobia);
    Metabolism: rarely - decrease or increase in body weight;
    Allergic reactions: rarely - allergic dermatitis, rash, vasomotor disorders, urticaria;
    Dermatological reactions: rarely - alopecia;
    Others: a feeling of rush of blood to the face, accompanied by a feeling of heat (disappears after the end of therapy).

Overdose

An overdose of clomiphene usually manifests itself with the following symptoms: a feeling of flushed face, nausea, vomiting, visual disturbances (flashes, decreased visual acuity, scotoma), vasomotor reactions, pain in the abdomen and pelvic organs, enlargement of the ovaries.
Conducting symptomatic and supportive therapy is shown. There is no information available on the effectiveness of dialysis to eliminate clomiphene.


Special instructions
