Release form and composition

Dosage form of Lasolvan Rhino is a nasal dosage spray: a clear solution of pale yellow color with the smell of eucalyptus (10 ml each in glass brown bottles equipped with a metering device and a nasal adapter with a protective cap, 1 bottle in a cardboard box).
Composition of 1 dose of the drug:
    active substance: tramazoline hydrochloride monohydrate - 82 μg (in terms of tramazoline hydrochloride);
    additional components: purified water - 66 358 mcg, povidone - 2101 mcg, glycerol 85% - 700 mcg, citric acid monohydrate - 270 mcg, sodium chloride - 183 mcg, sodium hydroxide - 154 mcg, magnesium sulfate heptahydrate - 49 mcg, hypromellose ( hydroxypropyl methylcellulose) - 35 μg, magnesium chloride hexahydrate - 35 μg, L-menthol (levomenthol) - 14 μg, benzalkonium chloride - 14 μg, racemic camphor - 14 μg, calcium chloride dihydrate - 11 μg, cineole (eucalyptol) - 7 μg, sodium bicarbonate - 1 mcg.

Pharmacodynamics

Tramazolin hydrochloride - the active substance of Lasolvan Rhino - is an alpha2-adrenergic agonist that has a vasoconstrictor effect. Due to the narrowing of the vessels after getting on the mucous membranes of the nose, the drug reduces their swelling, as a result of which the patency of the nasal passages is quickly restored and nasal breathing is facilitated.
The drug begins to act 5 minutes after instillation, its effect persists for 8-10 hours.

Pharmacokinetics

The pharmacokinetics of tramazoline in humans has not been studied. During studies on monkeys, rabbits and rats, it was found that when using the drug, 50 to 80% of the dose is absorbed orally or intranasally. Both tramazolin and its metabolites are distributed in all internal organs; the highest concentration with constancy was found in the liver.
The main metabolites of tramazoline after oral and topical administration are determined in the urine.
The elimination half-life makes 5–7 hours.

Indications for use

According to the instructions, Lasolvan Rhino is prescribed to relieve swelling of the mucous membrane and eliminate nasal congestion caused by acute respiratory infections and / or hay fever (rhinitis, hay fever).

Contraindications

Lasolvan Rhino is applied intranasally.
Adults and children from 6 years of age are prescribed 1 injection into each nostril. The frequency of instillations should not exceed 4 times a day. Before injection, it is recommended to clean the nasal passages.
The duration of treatment is 5-7 days.
Use of the drug:
    Remove the protective cap from the bottle.
    Before first use, press the metering device a few times into the air until a steady cloud of aerosol appears.

Overdose

With an overdose of tramazoline, blood pressure (BP) can increase and tachycardia can occur, followed by them, especially in children, a sharp decrease in blood pressure, lower body temperature, the development of reflex bradycardia and shock are possible.
Considering the experience of using other alpha-adrenergic agonists, the clinical picture of intoxication may be unclear, due to the probability of a change in the phases of stimulation and depression of the central nervous system (CNS) and the cardiovascular system (CVS), especially in children. Symptoms of central nervous system stimulation: agitation, anxiety, hallucinations, convulsions. Symptoms of central nervous system depression: lethargy, decreased body temperature, drowsiness, coma.
Also, in case of an overdose, the following symptoms may develop: respiratory failure (including respiratory failure, respiratory arrest), cyanosis of the lips, myosis, mydriasis, pallor, fever, sweating, impaired CCC functions (including bradycardia, cardiac arrest ), mental disorders.
In case of a nasal overdose of tramazoline, it is necessary to rinse the nose immediately. When symptoms of intoxication appear, symptomatic therapy is performed.
